摘要
目的:探讨自主吸入不同浓度七氟烷(sev)对正常大鼠肺泡上皮液体清除及肺泡毛细血管通透性的影响。方法:70只Wistar大鼠随机取出7只设为C组(正常组,吸入空气)。剩余大鼠随机分为三组,每组21只:O组(吸入40%氧气)、S1组(吸入1.5%七氟烷)、S2组(吸入3.0%七氟烷),每组根据吸入时间不同又分为三个亚组:4、8和10 h,每亚组7只大鼠,S1组和S2组的10 h亚组是吸入七氟烷8 h后停药,再吸入40%氧气2 h,所有组的新鲜气体流量为3.5 L/min。实验前给大鼠经尾静脉注射伊万斯兰50 mg/kg。于停止吸入气体时处死大鼠取肺组织测定湿/干重比、肺水含量以及提取肺组织中伊万斯兰来测定肺泡毛细血管膜通透性。结果:氧气组与空气组相比肺组织湿/干重比、肺水含量和伊万斯兰含量/肺组织重量之比无统计学差异(P>0.05),吸入相同时间不同浓度的七氟烷与吸入相同时间的氧气组的各亚组相比肺组织湿/干重比、肺水含量和伊万斯兰量/肺组织重量的差异无统计学意义(P>0.05)。结论:吸入长时间8 h高浓度(3%)七氟烷并不会对正常大鼠肺脏的肺泡上皮液体清除及肺泡毛细血管通透性产生影响。
Objective To investigate the effects of spontaneously inhaled different concentrations of sevoflurane on the clearance of pulmonary edema fluid and the membrane permeability of alveolar capillaries in normal rats. Method 70 Wistar rats were randomly divided into group C, O, S1 and $2 after injection of Evans blue (50 mg/kg). Control group (Group C, n = 7) breathing room air; Oxygen group breathing 40% 02 (Group O, n = 21), S1 and $2 group breathing 1.5% and 3.0% sevoflurane in 40% 02 respectively (Group S1, S2,n = 21). Group O, was further divided into 3 subgroups according to the duration of 40% 02 inhalation for 4 h, 8 h and 10 h (n = 7 each). Group S1 and $2 were further divided into 3 subgroups (n=7, each) breathing sevoflurane for 4 h, 8 h and 8 h followed by 2-h 02 (40%) inhalation. The animals were sacrificed at the end of 02 or/and sevoflurane inhalation. Lung wet/dry weight ratio and lung water content were determined. The permeability of alveolar capillaries was determined by detecting the content of Evans blue. Result There was no significant difference in wet/dry weight ratio, lung water content and Evans-blue-content/lung weight ratio between group C and group O (P 〉 0.05). The group with inhalation 1.5% or 3% sevoflurane did not produce any significant effect on the lungs as compared to the group O (P〉0.05). Conclusion Inhalation of high concentration (3%) of sevoflurane for long duration (8 h) did not alter the clearance of pulmonary edema fluid and the alveolo-capillary barrier function in normal spontaneous rats.
