实验性低血糖对糖尿病大鼠血清内皮素-1的影响 被引量:8

Effect of hypoglycemia on serum endothelin 1 of diabetic rats

摘要 目的探讨实验性低血糖和血糖波动对糖尿病大鼠血清内皮素-1(ET-1)的影响。方法将正常Wistar大鼠随机分为对照(NC)组和低血糖(NL)组,糖尿病大鼠随机分为对照(DC)组、单纯低血糖(DL)组和低血糖后高血糖(DLH)组。用胰岛素诱导低血糖。DLH组于低血糖后给予50%葡萄糖灌胃诱发高血糖。所有实验大鼠于胰岛素或生理盐水注射前后1.5、4.5、6.5h测定血糖和ET-1。结果低血糖各组血清ET-1水平明显升高(DLH组>DL组>NL组);NL组ET-1水平在血糖最低点时最高(116.83±11.68)pg/ml;DL组和DLH组ET-1水平在血糖最低点时均无升高,峰值呈延迟现象;DLH组血糖再度升高时ET-1水平开始升高,峰值高于DL组[(239.3±22.22)pg/ml vs(161.12±55.94)pg/ml,P<0.05]。结论低血糖对糖尿病大鼠血管内皮损伤更明显;血糖剧烈波动对血管内皮损伤比持续低血糖更严重。 Objective To ascertain the influence of insulin-induced acute hypoglycemia and glucose fluctuation an the serum endothelin 1 (ET-1) of diabetic rats. Methods Healthy male Wistar rats were randomized into control group (NC) and hypoglycemia group (NL). Diabetic male Wistar mice were randomized into control group (DC), hypoglycemia group (DL), and hypoglycemia {oUowed by hyperglycemia group (DLH). Hypoglycemia was induced by intraperitoneal administration of insulin. Hyperglycemia was induced in the DLH group by intragastric administration of 50% glucose solution. Blood samples were obtained before and 1.5 h, 4. 5 h, and 6.5 h after injection to detect blood glucose and ET-1. Results The level of ET-1 in each hypoglycemic group was increased significantly (DLH〉DL〉 NL). ET-1 reached the maximum when the blood glucose in the NL group reached the minimum (116.83 ±11.68) pg/ml. ET-1 didn't increase in the DL and DLH groups when the blood glucose reached the minimum, with a delayed presence of its peak. ET-1 began to increase when the blood glucose was increased again in the DLH group, with a significantly higher peak than in the DL group [(239. 39±22. 22) pg/ml vs (161.12±55. 94) pg/ml (P〈0. 05)]. Conclusion Hypoglycemia causes more serious injury of endothelium ceils of diabetic rats, and glucose fluctuation induces more serious injury of endothelium cells than uncontrolled hypoglycemia.
