银杏叶提取物对肝纤维化大鼠肝组织COX-2表达的影响 被引量:7

Effects of Ginkgo biloba Extracts(GbE) on Cyclooxygenase-2 in Liver of Rats with Hepatic Fibrosis

摘要 目的:观察银杏叶提取物(GbE)对肝纤维大鼠肝组织环氧合酶-2(COX-2)、Ⅰ型胶原表达的影响,探讨其抗肝纤维化的作用机制。方法:采用40%的CCl4皮下注射6周建立大鼠肝纤维化模型,以200、100、50mg.kg-1.d-1的GbE干预,原位杂交检测肝组织COX-2mRNA表达,免疫组化检测肝组织Ⅰ型胶原、COX-2蛋白表达。结果:模型组大鼠肝组织Ⅰ型胶原蛋白、COX-2mRNA和蛋白表达均较正常组明显增强,应用GbE干预后,大鼠Ⅰ型蛋白、COX-2mRNA和蛋白表达均明显减少。结论:GbE抗肝纤维化作用的机制之一可能与其下调肝组织中COX-2的表达,减少胶原的产生,改善肝脏微循环有关。 Objective:To evaluate the effectiveness of the Ginkgo biloba Extracts (GbE) on rats with hepatic fibrosis and the influence on cyelooxygenase -2 (COX -2) and typeI collagen protein expression and to explore its anti -fibrosis mech- anism. Methods : Hepatic fibrosis of rats were induced by carbon tetraehloride, then all the rats were respectively given with 200 mg/kg, 100 mg/kg, 50 mg/kg of GbE by daily gavage. The expression of COX - 2 mRNA was detected by fluorescent in situ hybridization. The protein expressions of COX - 2 and typeIcollagen were detected by immunohistochemistry. Results : The hepatic typeIcollagen protein and the mRNA and protein expressions of hepatic COX -2 expression of the rats in the model group were higher than those in the normal control group and the hepatic typeIco]lagen protein and the mRNA and protein expressions of hepatic COX - 2 expression of the rats in the GbE groups were significantly reduced. Conclusions : GbE could obviously inhibit the expression of COX - 2, reduce collagen production and improve liver micro circulation, and it may be one important mechanism for GbE to treat hepatic fibrosis.
