摘要
目的:探讨蟾皮对肾癌细胞GRC-1凋亡的诱导作用及Fas、Fas配体(Fas Linand,FasL)和bcl-2表达的影响,了解其抑制肿瘤的分子生物学机制。方法:用末端脱氧核苷酰转移酶法(TdT-mediated dUTP nick end labeling,TINEL)流式细胞技术检测细胞凋亡,免疫细胞化学检测Fas、FasL和bcl-2的表达。结果:当蟾皮提取液浓度分别为0μg/ml,0.01μg/ml,0.1μg/ml,1μg/ml,10μg/ml时,GRC-1的凋亡百分比分别为0.97%,1.15%,1.02%,7.06%,20.32%。随着药物浓度的增加,Fas表达增强,FasL和bcl-2表达减弱。结论:蟾皮抑制肾癌细胞的分子生物学机制之一为促进Fas表达,抑制FasL及bcl-2的表达,诱导癌细胞凋亡。
Purpose: : To study the effect on apoptosis induced by the extract from toad skin arid expression of Fas, Fas Ligand (FasL) and bcl - 2 in renal cell lines (GRC - 1), and the molecular mechanisms of anti - cancer of toad skin. Methods: : Apoptosis was test by TdT - mediated dUTP nick end labeling (TUNEL). The expression of Fas, FasL and bcl -2 was determined by imraunohistochemical staining. Results: When the concentration of the extract was 0μg/ml, 0.01 μg/ mL, 0.1μg/ml.1μg/ml, 10μg/ml, the apoptosis percentages of renal cell lines was 0. 97%, 1.15%, 1.02%, 7.06%, 20. 32% respectively. And the expression of Fas increased, FasL and bcl - 2 decreased with the concentration of the extract increasiag. Conclusion: One of the molecular mechanisms of anti - cancer of toad skin in renal cell lines is to induce apoptosis and increase the expression of Fas, decrease the expression of FasL and bcl - 2.
