摘要
洋甘菊因含有丰富生物活性成分,具有多种保健作用而受到人们广泛关注,其中芹菜苷(Apioside)是其主要活性物质。本实验建立了HPLC测定洋甘菊中芹菜苷含量的方法,采用有机溶剂回流法提取洋甘菊中的芹菜苷,并用HPLC测定芹菜苷的含量。在单因素试验的基础上,采用响应面分析法对洋甘菊提取芹菜苷提取条件进行优化。确立了提取的最佳工艺条件:乙醇体积分数为67%,料液比是1∶24,提取温度为72℃,提取时间为2 h。试验证明,此条件下芹菜苷率为2.14%(n=3),与模型预测2.096%基本一致。
The objective of this study was to develop an HPLC method for determination of apioside in chamomile. The extraction condition was optimized using response surface analysis based on single factor experiments. The model was es- tablished and analyzed by response surface methodology (RSM). The optimal reflux extraction condition was established as follows:the solvent was alcohol, the concentration of alcohol was 67%, the material/solvent ratio was 1: 24, the tem- perature was 72 ℃ ,the time was 120 minutes. The content of apioside extracted out was determined by HPLC. The ex- traction ratio of apioside was determined to be 2.14% (n =3) using the optimized extraction condition,which was close to the estimated value from model.
