摘要
目的:研究二苯乙烯苷(2,3,4’,5-tetrahydroxystilbene-2-O-β-D-glucoside,TSG)对精氨酸加压素(argininevasopressin,AVP)引起的心肌成纤维细胞(cardiac fibroblasts,CFs)增殖的抑制作用及其相关信号通路。方法:采用MTT法观察TSG对AVP引起的CFs增殖的抑制作用。定磷法检测细胞内钙调神经磷酸酶(calcineurin,CaN)的活性,WesternBlot测定其蛋白表达。结果:30μmol/L、100μmol/L的TSG明显抑制100 nmol/L AVP引起的CFs的增殖(P<0.01),而3μmol/L、10μmol/L的TSG则对CFs的增殖无明显影响。CaN的活性在100 nmol/L AVP刺激30 min后达到最高(P<0.05),而30μmol/L TSG能抑制CaN活性的增高(P<0.05);CaN蛋白水平在AVP刺激48 h后明显升高(P<0.05),而这种升高作用能被30μmol/L TSG抑制(P<0.05)。结论:TSG可以抑制AVP诱导的CFs的增殖。TSG可能是通过减少CaN活化和蛋白表达抑制CFs的增殖。
Objective: To evaluate the effects of 2,3,4',5-tetrahydroxystilbene-2-O-β-D-glucoside(TSG) on arginine vasopressin(AVP)-induced cardiac fibroblast(CFs) proliferation and to explore the underlying intracellular mechanisms.Methods: CF proliferation was assessed by a cell survival assay MTT.CaN activity and protein levels were determined by activity assay kit and Western Blot.Results: TSG(30-100 μmol/L) significantly inhibited AVP(100 nmol/L)-induced proliferation of rat neonatal CFs(P〈0.01),while TSG(3-10 μmol/L) had no obvious effect.CaN was activated by AVP and peaked at 30 min(P〈0.05),which was inhibited by TSG(P〈0.05).CaN protein level was markedly increased after stimulation with AVP for 48 h(P〈0.05),whereas 30 μmol/L TSG could inhibit this kind of effect(P〈0.05).Conclusion: TSG inhibits CaN activation and expression,and consequently suppresses cell proliferation induced by AVP.
