摘要
[目的]探讨SD大鼠膝内侧副韧带(medial collateral ligament,MCL)损伤后早期跑台运动对胶原纤维形成的影响。[方法]健康成年雄性SD大鼠36只,随机分为实验组和对照组,每组18只,制备大鼠双膝MCL断裂动物模型。实验组每天进行一次定时定量的跑台运动;对照组置于饲养笼内不做运动干预。术后每周两组分别处死3只大鼠,采集双下肢MCL损伤处韧带组织做标本,分别行HE染色和Masson's染色,进行组织学观察。采用Im-agepro plus 6.0图像分析软件分别计算胶原纤维面积及平均光密度值。[结果]伤后第1周时实验组较对照组韧带损伤处间隙增宽,第6周时实验组较对照组韧带损伤处瘢痕体积增大,排列不规则;HE染色显示实验组伤后各周损伤局部的炎性反应过程均较对照组强烈,且胶原纤维排列较对照组紊乱;Masson's染色显示随着伤后时间的延长,除第1周两组比较无显著性差异外(P>0.05),其余各周对照组的胶原纤维面积依次为54.4%、60.7%、68.0%、76.5%、80.7%,均显著高于实验组相对应的51.4%、56.5%、59.3%、67.1%、71.1%(P<0.01);平均光密度值的变化与Masson's染色的变化趋势一致。[结论]SD大鼠膝内侧副韧带损伤后早期运动训练可减慢新生胶原纤维组织的形成,导致韧带延迟愈合,对韧带损伤后的修复造成负面影响。本实验结果对指导运动人群韧带损伤的康复具有一定指导意义。
[ Objective] To explore the effects of early treadmill exercise on formation of collagen fibers in an animal model of medial collateral ligament (MCL) injury. [ Method] Thirty six healthy adult male SD -rats were randomly divided into the experimental group and the control group,with 18 rats in each group. MCLs on both knees were cut off transversly to create a MCL injury model. The animals in experimental group underwent quantitative treadmill exercise once daily, by contrast, the rats in control group were breed in the cage without any movement intervention. Every week after operation,3 rats in each group were killed respectively histological for study. The double lower limbslnjury ligaments of MCL of these rats were collected as speci- mens,then the two groups of specimens underwent HE staining and Masson's staining respectively to make histologic observa- tion. The area of collagen fiber and the average optical density were calculated by Imagepro plus 6 image analysis software. [ Re- suit] The space of ligamentous injury in experimental group was wider than that in the control group in the first week. The vol- ume of ligamentous injury in experimental group was bigger than that in the control group in the sixth week, and the arrangement was more irregular. The HE staining showed that inflammatory reaction process of experimental group was stronger than that in the control group every week after injury, and collagenous fiber was more disordered than that in the control group. The Masson' s staining showed that with the extension of time after injury, except for the first week there were no significant difference be- tween the collagen fiber area of the control group and the experimental group (P 〉 0.05), the other weeks showed significant differences (P 〈 0.01 ). For the experimental group, the collagen fiber area from the second to sixth week were successively 5 I.4% ,56.5% ,59.3% ,67.1% ,71.1% ,while for the control group, they were 54.4% ,60.7% ,68.0% ,76.5% ,80.7% ,which were obviously higher than the experimental group. [ Conclusion ] The early training of SD - rats after MCL injury will slow down the for- mation of new collagen fibers, resulting to delayed ligaments heal- ing, thus producing negative effects on the repair of medial collateral ligament. This experiment has certain significance in guiding the re-habilitation of sports ligament injury.
