高压氧治疗对大鼠肝脏缺血再灌注损伤的保护作用

Protective effect of hyperbaric oxygen therapy on liver ischemic reperfusion in rats

摘要 目的探索高压氧(HBO)对肝脏缺血再灌注(IR)损伤的作用及其干预方法。方法健康雄性SD大鼠40只,随机分为假手术组(SO)、缺血再灌注组(IR)、高压氧治疗1次组(HBO1)、高压氧治疗2次组(HBO2)。观察动物苏醒时间;术后5日再次开腹对比肝脏大体观;检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平;取活体肝脏组织行光学显微镜观察。结果 HBO治疗组术后清醒快,出舱后均清醒;HBO治疗组的肝脏瘀血、水肿程度较轻;HBO治疗1次组的ALT、AST均值分别为395.52、431.75 U/L,与IR组比较,差异有统计学意义(P<0.05);光学显微镜观察结果:HBO治疗组的肝细胞水肿程度最轻,IR组最重。结论 HBO治疗肝脏IR损伤具有保护作用,HBO每日治疗1次与2次比较效果相同。 [ Objective ] To explore the effect of hyperbaric oxygen therapy { HBO) on liver ischemic reperfusion ( IR} , and its inter- vention method. [ Methods ] 40 male SD ( Sprague-Dawley } rats were randomly divided into 4 groups : sham operation group { SO group} , isehemia reperfusion group ( IR group} , once hyperbaric oxygen therapy group ( HBO1 group} , and twice hyperbaric oxygen therapy group ( HBO2 group ). The revive time was observed. The view of the liver was compared 5 days after the operation. The levels of serum alanine transaminase { ALT) and aspartate transaminase (AST) were detected. The hepatic tissue was observed with light microscope. [ Results] The rats in HBO groups were revived quickly than other groups, and all rats were awake. The de- grees of liver blood stasis and edema were slight in HBO groups. The level of ALT and AST in HBO1 group was 395.52 U/L and 431.75 U/L respectively, which the differences in levels of ALT and AST between HBO1 group and IR group were significant { P 〈 0.05 }. Under the light microscope ~ the mildest edema of liver cells was seen in HBO groups but most serious in IR group. [ Conclusion] Hyperbaric oxygen therapy has a protecting effect on liver isehemic reperfusion injury, and the effect of therapy once daily is the same as that of therapy twice daily.
