摘要
采用Plackett-Burman(PB)试验、最陡爬坡试验、响应面设计法对双水相萃取分离葡萄籽多糖的条件进行优化。首先,采用Plackett-Burman设计从影响萃取率的5因素中筛选出PEG质量分数、硫酸铵质量分数和萃取温度3个显著影响因素。在此基础上,通过最陡爬坡试验逼近最大萃取率区域,然后通过响应面组合试验对显著因素进行优化。得到最佳萃取条件为:PEG2000质量分数为8.92%、(NH4)2SO4质量分数为16.41%、萃取温度为38℃,萃取率可达55.1%,与预测值56.075%接近。
Extraction and separation of polysaccharide from grape seeds by two-phase aqueous systems was investigated by Plackett Burman(PB) de- sign, steepest ascent design and response surface methodology. Firstly, PB design was applied to identify the mass fraction of PEG2000 and (NH4)2SO4 and extraction temperature as the most significant of 5 factors influencing the extraction. Then the center points of the three factors were determined based on steepest ascent design. Then the optimization of the three factors was carried out by using response surface methodology. The results showed that the best extract conditions were as follows: the mass fractions of PEG2000 8.92%, the mass fractions of (NI-L)2SO4 16.41%, temperature at 38℃. Under such conditions, the extraction rate was 55.1%, which was in good agreement with the predicted one of 56.075%.
