EPA和DHA对脂多糖刺激大鼠系膜细胞β1整合素和整合素连接激酶表达的影响 被引量:1

摘要 目的观察二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)对脂多糖(LPS)刺激的大鼠系膜细胞(GMCs)β1整合素(β1 integrin)和整合素连接激酶(integrin-linked kinase,ILK)表达的影响。方法用LPS(10mg/L)刺激大鼠GMCs,经EPA(10μmol/L、100μmol/L)、DHA(10μmol/L、100μmol/L)培养24 h、48 h后,采用实时定量PCR方法检测β1 integrin和ILK mRNA的表达。结果 LPS刺激可使GMCsβ1 integrin和ILK mRNA的表达增加;经EPA和DHA处理后,β1 integrin和ILK mRNA的表达下降。结论 EPA和DHA可以通过降低LPS诱导的GMCsβ1整合素和ILK的表达,对损伤的GMCs起保护作用。 Objective: To observe the effects of eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) on lipopolysaccharide(LPS) induced expression of β1 integrin and integrin linked kinase(ILK).Methods:The glomerular mesangial cells(GMCs) were stimulated by lipopolysaccharide(LPS) and incubated with EPA(10 μmol/L or 100 μmol/L) and DHA(10 μmol/L or 100 μmol/L) for 24 h or 48 h.The mRNA expressions of β1 integrin and ILK were detected by real-time PCR method.Results:LPS promoted β1 integrin and ILK mRNA expression,while EPA and DHA inhibited the GMCs β1 integrin and ILK mRNA expression induced by LPS.Conclusion:EPA and DHA might inhibit the GMCs β1 integrin and ILK mRNA expression induced by LPS,for preserving injury GMCs.
