摘要
为探究蒙古黄芪2种表型种子在遗传上的分化情况,采用垂直板聚丙烯酰胺凝胶电泳技术,对蒙古黄芪有纹和无纹2种表型种子共90粒个体的酯酶同工酶进行了研究。结果表明:电泳共产生的27条酯酶同工酶谱带中,分别有11.11%、29.63%和70.37%的谱带在种子个体、母株个体及种子表型水平上是恒定的,与其相应的其余谱带分别在上述3个水平上存在不同程度的差异。聚类分析将90粒供试种子划分为2个酶谱表征群和6个亚表征群,其中B表征群均由有纹种子组成,A表征群则由有纹和无纹2种表型种子组成;除c亚表征群由2种表型种子组成外,其余5个亚表征群均由同一表型的种子组成。蒙古黄芪2种表型种子在遗传上未明显分化,表型和遗传上并不完全一致,需进一步加强人工选择。
Vertical polyacrylamide gel electrophoresis was used to study the esterase isozyme system in the 90 seed individuals to uncover the genetic relationship between two seed phenotypes of Astragalus membranaceus var.mongholicus,with one type possessing spots on the surface of seed coat but the other not.The results showed that a total of 27 esterase isozymic bands were generated,of which 11.11%,29.63%,and 70.37% separately existed stably at seeds,mother plants,and seed phenotypes levels,while 88.89%,70.37% and 29.63% were ploymorphic among above-mentioned three levels,respectively.In the dendrogram,all the seeds tested were divided into two groups or six subgroups.Group B only included seeds with spots,while group A had two phenotypic seeds.Among the six subgroups,subgroup c possessed two phenotypic seeds and the others consisted of the same phenotypic seeds.In conclusion,there was no obvious genetic differentiation between the two seed phenotypes of A.membranaceus var.mongholicus,but futher artifical selection could be helpful to accelerate genetic differention between them.
