摘要
目的:分离、鉴定海地瓜中的一脂类化合物,测定其体外抗肿瘤活性。方法:对海地瓜干粉用氯仿-甲醇-水体系(体积比4∶4∶1)萃取,经连续正反相柱层析分离得到1种脂类化合物。采用IR、ESI-MS、NMR、GC-MS等分析手段对该化合物结构进行表征,并用MTT法检测其对S-180细胞增殖的影响。结果表明:该活性脂质为神经酰胺类化合物系列物,主要由2-氨基-1,3-二羟基-15-甲基-4-十七烯长链碱和5种饱和及单不饱和的脂肪酸构成。MTT试验表明其对S-180小鼠肉瘤细胞的增殖具有较好抑制活性,表现出显著的时间-剂量依赖效应,72h时的IC50值为172.45μg/mL。
Objective: Isolated and identificated a lipid component in Acaudina molpadioides, also carried on the vitro antitumor activity experiment. Method: The dry powder of A caudina molpadioides was extracted with chloroform/ methanol/water (4:4:1, V/V/V), then it was separated on silica-gel column chromatography and a purified lipid were ob- tained. The structure was identified by spectroscopy and chemical methods such as IR, ESI-MS, NMR and GC-MS. S- 180 cells were treated with ceramide and vitro antitumor activity was detected by MTF. Conclusion: The component is a series of ceramide compounds, which mainly consisted of 2-amino-l,3-dyhydroxy-15-methyl-4-heptadecene as long chain base and saturated and monounsaturated fatty acids. MT'F experiment showed eeramide can inhibit the proliferation activity of S-180 sarcoma cells in mice, and the effect was in an obvious time-and-dose-dependent manner. Its IC50 was 172.45μg/mL after 72 h.
