摘要
目的 探讨纳洛酮对急性心肌缺血面积的影响。方法 结扎冠状动脉左室支中点复制心肌缺血模型。采用纳洛酮干预 ,缺血 3个小时后静脉注射美兰使非缺血区兰染 ,处死动物分离左心室 (LV)并切片 ,用TTC染色 ,区分缺血心肌(IS)、危险心肌 (AAR)及正常心肌。在解剖显微镜下分离IS、AAR ,分别称重 ,观察IS、AAR、IS/AAR %及IS/LV %以评价缺血范围。结果 纳洛酮实验组IS、AAR、IS/LV %、IS/AAR %明显减少与对照组相比差异显著。结论 纳洛酮能明显缩小急性心肌缺血的缺血面积。其机理可能与改善心肌微循环、促进组织氧合、降低组织乳酸浓度、减少缺血区水肿、防止膜脂质过氧化。
Objective To study naloxone′s effects to infarct size of rabbit myocardiac ischemia.Methods Rabbit myocardiac ischemiac mode was reconstructed by ligating left ventricular branch of coronary atery.Experimental group was treated by naloxone.Patent blue dye was injected into the venous to stain the normally perfused region of the heart after myocardiac ischemia for three hours.After executed rabbits,left ventricular was separated,sliced and dyed by TTC(2,3,5-triphenyltetrazonlium chloride)to differentiate infarct size(IS),area at risk(AAR)and normal myocardial.The fixed tissue piece were aparted under anatomy scope and then weighted.IS,AAR,IS/AAR% and IS/LV% were observed to assess infarct size.Results Above indexes of experimental group is smaller than that of contral group with famously statistic difference.Conclusion Naloxone can obviously reduce infarct size of myocardiac ischemia.Its mechanism probably have something to do with improving microcirculation,increasing oxygenation,decreasing latic acid concentration,reducing idema of ischemic myocardial and preventing lipid peroxidation and secondary lesion of ischemia [
