摘要
目的:建立金银花指纹图谱混合酸流动相测定方法。方法:采用高效液相色谱法,色谱柱为DIKMADiamon-silC18(2)色谱柱(4.6mm×250mm,5μm),以乙腈为流动相A,以含0.025%磷酸、0.3%醋酸的溶液为流动相B,进行梯度洗脱;检测波长为243nm。柱温为40℃。结果:使用混合酸能改善30~50min各色谱峰的分离度,使52min混合峰完全基线分离。结论:混合酸流动相使金银花色谱峰分离度增加,可用于金银花指纹图谱的研究。
Objective : To establish a HPLC mixed acid mobile phase method for determination of the fingerprint of Lonicera ja-ponica Thunb. Methods :A Diamonsil C18 (2)column(4.6 mm × 250 mm, 5 μm) was used, acetonitrile as the mobile phase A, a mixed solution of 0. 025% phosphoric acid,0. 3% acetic acid as the mobile phase B, gradient elution, detective wavelength was 243 nm and column temperature was 40 ℃. Results : The mixed acid mobile phase can improve separation 30 to 50 minutes chrom-atographic perks, making the mixed peaks of 52 minutes complete baseline separation. Conclusion:The mixed acid mobile phase can increase separation and suitability for the determination of the fingerprint of Lonicera japonica Thunb.
