摘要
目的建立HPLC法测定苦参素中氧化苦参碱的含量测定方法。方法采用PLATISIL ODS-C18色谱柱(250 mm×4.6 mm,5μm),流动相为磷酸二氢钾溶液(0.05 mol.L-1)-甲醇(83∶17),用磷酸调节pH值至3.5,流速为1.0 mL.min-1,检测波长为215 nm,柱温为30℃。结果氧化苦参碱的线性范围为18.98~189.84μg.mL-1(r=0.999 9),平均回收率(n=9)为96.2%,RSD为2.2%。结论 HPLC法测定苦参素中氧化苦参碱的含量准确、可靠,专属性强、灵敏度高,适用于苦参素中氧化苦参碱的含量测定。
Objective To establish a method for the marine determination oxymatrine contents by HPLC. Methods The chroma- tographic condition was as follows: PLATISIL ODS- C18 (250mm×4.6mm,5μm) column was adopted, the mobile phase was composed of potassium dihydrogen phosphate solution (0.05 mol/L) -methanol (83:17 ), and adjusting to pH 3.5 with phosphoric acid, the flow rate was 1.0 ml/ min, the detection wavelength was at 215nm and the column temperature was 30℃. Results The calibration curve showed a good linearity in the range of 18.98 - 189.84 μg/ml(r =0. 9999). The average recovery (n =9) was 96.2% and RSD was 2.2%. Conclusion The method is accurate, reliable, stable and sensitive, and it can be applied for the oxymatrine contents of marine.
