抗纤灵有效单体对活化的肾成纤维细胞株和系膜细胞株的影响 被引量:2

摘要 目的:观察抗纤灵有效单体对活化的肾成纤维细胞株和系膜细胞株的影响。方法:采用体外培养肾成纤维细胞株和系膜细胞株的方法,将其分为空白对照组、TGF-β_1 2ng/mL组、TGF-β_12ng/mL+中药10^(-5)g/mL组和TGF-β_12ng/mL+中药10^(-6)g/mL组。采用MTT法观察24h中药单体对两种细胞株的作用以及ELISA法对TGF-β的分泌情况。结果:与空白对照组相比,加入TGF-β_1刺激因子可以促进两种细胞株的活化(P<0.05),以及TGF-β的分泌(P<0.01),中药单体可以抑制活化的细胞株增殖和TGF-β的分泌(P<0.05,P<0.01)。结论:抗纤灵中药单体可以抑制活化的肾成纤维细胞株和系膜细胞株增殖,有效防治肾纤维化,其作用机制可能与其抑制TGF-β的分泌有关。 Objective: To observe the effect of utility monomer in Kangxianling to the renal fibroblast cell line and mesangial cell line. Methods: Adopt the method of culturing the renal fibroblast cell line and mesangial cell line in vitro, which were divided into blank control group, TGF-β1 2ng/mL group, TGF-β1 2ng/mL+Chinese medicine monomer 10-5g/mL group and TGF-β1 2ng/mL +Chinese medicine monomer 10 6g/mL group. Observing the effect of Chinese medicine monomer for 24 hours to the two cell lines by MTT assay, and the secretion of TGF-β by ELISA assay. Results: Compared with blank control group, adding TGF-β1-stimulating factor could promote the two cell lines (P〈0.05), as well as the secretion of TGF-β (P〈0.01), Chinese medicine monomer could inhibit the activation of two cell lines proliferation and secretion of TGF-β (P〈0.05, P〈0.01). Conclusion: Utility monomer of Kangxianling can inhibit the activation of the renal fibroblast cell line and mesangial cell line proliferation and secretion of TGF-β, effectively prevent and treat renal fibrosis, and its possible mechanism of action is related to inhibition of TGF-β secretion.
