氟中毒大鼠生长板Ihh、PTHrP mRNA及蛋白表达的变化 被引量:4

Effect of fluoride on mRNA and protein expression of Ihh and PTHrP in bone growth plate of rat

摘要 目的观察氟中毒大鼠生长板Ihh、PTHrP mRNA及蛋白表达的影响,探讨Ihh信号通路在氟致软骨损伤中的可能作用。方法 32只Wistar大鼠按体重随机分成4组,对照组饮用自来水,染氟组分别饮用含氟50、100和150 mg/L的自来水,3个月后测定大鼠血清及骨组织氟含量,Real-time PT-PCR法检测Ihh、PTHrP mRNA表达情况,免疫组化法检测软骨组织中PTHrP蛋白表达情况。结果染氟组大鼠生长板中Ihh、PTHrP mRNA表达显著高于对照组,差异有统计学意义(P<0.05)。对照组大鼠生长板软骨细胞不表达或极弱表达PThrP,而氟中毒大鼠生长板静止层和肥大层软骨细胞细胞质中PTHrP蛋白为阳性表达,且随染氟剂量增加而升高。结论过量氟可能通过影响Ihh、PTHrP mRNA及蛋白表达水平,造成生长板软骨增殖、分化失衡。 Objective To observe the effect of excess fluoride on mRNA and protein expression of Ihh and PTHrP in bone growth plates of rats, thereby further explore the role of Ihh signaling pathway in cartilage damage induced by excessive fluoride ingestion. Methods Thirty-two Wistar rats were randomly divided into four groups : control rats were only given tap water, the fluoride exposed rats were given fluoride containing tap water (50, 100 and 150 mg/L of fluorion), three months later, the content of fluoride in serum and bone was determined, and the Ihh, PTHrP mRNA expression was also measured by real-time RT-PCR, and PTHrP protein expression were detected by immunohistochemistry at the same time. Results The mRNA expression of Ihh and PTHrP in growth plates of fluoride-treated rats were significantly higher than those of control group ( P 〈 0. 05 ), immunohistochemistry showed that the PTHrP protein in growth plate of fluorosis rats was also significantly increased in ehondro- cytes of quiescent layer and hypertrophic layer than that of control rats. Conclusion Excess fluoride exposure may stimulate the expression of Ihh, PTHrP mRNA, resulted in imbalance of ehondrocytic proliferation and differentiation in bone growth plate.
