摘要 目的:探讨紫外分光光度仪在临床百草枯中毒中的应用,建立血清中百草枯定性定量检测的紫外分光光度法。方法:血清经20%三氯醋酸沉淀蛋白,-20℃冰箱放置10 min,12 000 r/min高速离心,取上清液,采用50μL微量比色池,紫外分光光度法测定。结果:血清中百草枯浓度在0.10~40μg/mL范围内呈线性关系,回归方程y=0.081 2x+0.010 4,其相关系数为0.999 6,回收率为92.5%~103.0%,相对标准偏差(RSD)为3.6%~4.7%。日内、日间RSD分别为2.8%~4.8%和2.2%~4.6%,最低检出限为0.05μg/mL。结论:该仪器和方法操作简便、分析快速、结果准确,为临床治疗疑似百草枯中毒患者的药物定性定量检测提供了一种简便准确的检测方法,适合对血液灌流前、后的百草枯浓度的测定。 To apply UV spectrophotometer for the diagnosis of paraquat poisoning and develop a method for qualitative and quantitative detection of serum paraquat by UV spectrophotometer. The serum had its protein precipitated with 20% TCA, and then went through 10-min storage in the -20℃ refrigerator and 12 000 r/min high-speed eentrifugation. The supernatant was measured by UV spectrophotometer, with 50μL microeell used. Linear correlation occurred when the serum paraquat concentration was limited between 0.10 μg/mL and 40 μg/mL, with the regression equation of y=0.081 2x+0.010 4, the coefficient correlation of 0.999 6, the recovery rate between 92.5% and 103.0%, relative standard deviation between(RSD) 3.6% and 4.7%. The RSD was between 2.8% and 4.8% in a day and between 2.2% and 4.6% in several days, with minimum detection limit of 0.05 μg/mL. A simple and accurate method is provided for the qualitative and quantitative detection of serum paraquat of the poisoning patient, which can be used for paraquat concentration measuring before and after hemoperfusion.
