摘要
以啤酒酵母(Saccharomycescerevisiae)I菌株为出发菌株,经紫外线处理后,在含2.0mg/L的5-(2-氨基乙基)-L一半胱氨酸(AEC)的平板上获得18株生长良好的抗性突变林,编号为I-1~18,其中Ⅰ-Ⅱ株抗AEC达10~15mg/L,是出发株抗性最高浓度的10倍左右,并且Ⅰ—11株的L-赖氨酸含量(重量与重量比,100%)较出发株提高3.89%。以Ⅰ-11株作亚硝酸诱变处理,在含30~120mg/LAEC的培养基上又获得18株抗性突变株,其中Ⅰ11-L和Ⅰ11-MAEC的耐受浓度可达1300mg/L,其赖氨酸的含量较Ⅰ菌株的提高了27.42%和40.28%。
18 resistant mutants were collected after treating Saccharomyces cerevisiae strain Ⅰ as their starting strain with Liltraviolet. They grew well in ndneral medium (MM) containing 2. 0μg/ml 5 - (2 -andnoethyl) - L - cysteine (AEC), in which strain Ⅰ could not grow. Ⅰ - 11, one of the mutants, could grow well in the MM containing 10 - 15μg/ml of AEC. About 10 times of maximum concentration that strain Ⅰ could resist. L - lysine in ferment broth of Ⅰ - 11 was 103. 98% of strain Ⅰ. Further treating Ⅰ -11with nitrite, another 18 strains were isolated from MM containing 30μg/ml of AEC. Two of thern, Ⅰ11 -L alld Ⅰ11 - M grew well in MM containing 1300μg/ml of AEC, and L - lysine conent in ferment juice of Ⅰ11 - L and Ⅰ11 - M were 127. 4 % and 140. 28% of strain I respectively.
