参附注射液对四氯化碳诱导肝纤维化肝组织TGF-β1和IL-1表达的影响 被引量:1

摘要 目的观察参附注射液对四氯化碳诱导大鼠肝纤维化过程中对转化生长因子β1(TGF-β1)及白细胞介素1(IL-1)表达的影响。方法 90只SD大鼠皮下注射四氯化碳连续7周,建立肝纤维化模型后,均分为3组。第8周开始药物治疗组分别给予参附注射液5 ml/kg和15 ml/kg。HE和Masson染色观察肝组织病理改变并进行肝纤维化分级;免疫组化法测定肝组织中TGF-β1及IL-1表达水平。结果参附注射液治疗组大鼠肝纤维化程度显著低于模型组(P<0.01),血清TGF-β1及IL-1表达水平也明显低于模型组(P<0.05)。结论参附注射液对四氯化碳诱导的大鼠肝纤维化具有显著的治疗作用,其作用机制可能与抑制TGF-β1、L-1表达有关。 Objective To investigate the effects of SENFU injecta on expressions of TGF-β1 and IL-1 in rat hepatic fibrosis induced by carbon tetrachloride (CC14). Methods The model of hepatic fibrosis was established by administration of CC14 to 90 SD rats two times a week for seven weeks. After that,the animals were divided into 3 groups that were treated with or without 5 ml/kg or 15 ml/kg SENFU injecta. HE and Masson stain were used to examine liver pathologic changes and grade of liver fibrosis. Immunohisto- chemistry was performed to detect the expressions of TGF-β1 and IL-1. Results The expressions of TGF-β1 and IL-1 were lower in the SENFU-treated rats than those in the untreated animals(P 〈 0. 05). The degree of liver fibrosis in SENFU-treated rat was signifi- cantly reduced when compared to the untreated animals(P 〈 0. 01). Conclusions The SENFU can significantly decrease the expres- sions of TGF-β1and IL-1 in hepatic tissue and inhibit liver fibrogenesis in rats
