摘要
采用同源克隆法结合cDNA末端快速扩增(RACE)技术克隆东北白桦(Betula platyphylla)咖啡酸/5-羟基阿魏酸-O-甲基转移酶(COMT)基因COMT1,测序结果显示东北白桦COMT1 cDNA序列全长1 367 bp(GenBank登录号:KC292201),其中完整编码区长度1 095 bp,编码365个氨基酸。利用Gateway技术分别构建了东北白桦COMT1植物过量表达和反义表达载体。
The caffeic acid-O-methyltransferase (COMT) gene COMT1 of Betula platyphylla was isolated by homology cloning method combing with rapid amplification of cDNA ends (RACE) technology. Sequening results showed that the total length of COMT1 cDNA segment was 1 367 bp(GenBank Accession Number: KC292201), of which the complete coding region was 1 095 bp, encoding 365 amino acids. Over expression vector and antisense expression vector of COMT1 were constructed through Gateway technique.
