Role of types A and C CpG oligodeoxynucleotide in nasal inflammation of allergic rhinitis mice

摘要 目的探讨A、C型CpG寡核苷酸(ODN)对卵清蛋白诱导小鼠鼻腔炎症反应的作用。方法 40只BALB/c小鼠随机分为正常对照组、变应性鼻炎组、A组和C组。变应性鼻炎组、A组和C组用卵清蛋白致敏及激发,A、C组在激发同时分别应用A、C型CpGODN干预,正常对照组用生理盐水做对照。观察各组小鼠鼻部症状评分、鼻黏膜病理改变及测量血清IL-4、IL-5和IFN?水平。结果 A、C组鼻部症状评分均高于正常对照组(P<0.05或0.01),但明显低于变应性鼻炎组(P<0.05或0.01)。变应性鼻炎组血清IL-4、IFN?水平最高,IL-5水平最低(P<0.01)。C组血清IL-4、IFN?水平最低,IL-5水平最高(P<0.05或0.01)。结论 A、C型CpGODN可抑制卵清蛋白诱导的小鼠鼻腔炎症,且C型CpGODN抑制作用更强。 Objective To observe the role of types A and C CpG oligodeoxynucleotide (ODN) in nasal inflammation of ovalbumin-induced allergic rhinitis (AR) mice. Methods Forty BALB/c mice were randomly divided into normal control, AR, A and C groups. AR, A and C groups were sensitized and challenged with ovalbumin, while normal controls were treated with normal saline. A and C groups received types A and C CpG ODN, respectively. Nasal symptom scores, pathology of nasal mucosa, and serum levels of IFN-γ, IL-4 and IL-5 were determined in all mice. Results Nasal symptom scores in A and C groups were higher than those in control group, but lower than those in AR group (P〈0.05 or 0.01). IL-4 and IFN-y levels were maximal in AR group but minimal in C group; while IL-5 content was maximal in C group but minimal in AR group (P〈0.05 or 0.01). Conclusion Types A and C CpG ODN can inhibit nasal inflammation in ovalbumin-induced AR mice, especially for type C CpG ODN.
