摘要
目的:确定大孔树脂纯化6-姜酚的最佳工艺条件。方法:以吸附量和解吸率为指标,筛选最佳树脂;以6-姜酚浓度为指标,确定最佳工艺参数。结果:HPD-950大孔树脂对6-姜酚纯化效果最好,最佳条件为:溶解粗提物乙醇体积分数35%,上样质量浓度2.1 mg/mL,pH 6.5,上样流速1 mL/min。当上样5.5倍柱体积时树脂达到动态饱和吸附,洗脱液乙醇体积分数65%,洗脱流速0.5 mL/min,洗脱液用量6倍柱体积,6-姜酚纯度达34.43%。结论:使用本工艺能简便、快速地分离纯化6-姜酚。
Objective: The present study aims at establishing an optimum method for purification of 6-gingerol by macroporous resin. Methods: The optimal macroporous resin was evaluated by indices including static absorption capacity and desorption rate, while the optimal technological parameter was determined by concentration of 6-gingerol. Results: Macroporous resin HPD-950 possessed the best purification property, and the optimum purification conditions were as follows: ethanol(which is used to dissolve crude extract) concentration 35%, initial sample solution concentration 2.1 mg/ mL, sample pH 6.0, flow rate 1 mL/min, dosage of 5.5 times column volume. And samples were desorbed by 65% ethanol with the amount of 6 times column volume, while elution rate was 0.5 mL/min. Under the mentioned conditions, purity of 6-gingerol reached 34.43%. Conclusion: As 6-gingerol could be separated in a simpler and faster way, the above processes are practical and feasible.
