选择70只小白鼠,雌雄各半,随机分为7组,其中第1组为对照组,口服生理盐水,第2~7组为试验组,第2~4组分别肌肉注射pcS/2SS、pGM-CSF/SS和pGM-CSF+pcS/2SS DNA疫苗,第5~7组分别口服以减毒沙门氏菌为载体的pcS/2SS、pGM-CSF/SS和pGM-CSF+pcS/2SS DNA疫苗,利用相对半定量RT-PCR检测小鼠肝脏组织生长激素受体(GHR)和胰岛素样生长因子-1(IGF-1)基因mRNA的表达。结果表明:3、5和6组的GHR mRNA表达高于1、2、4和7组(P<0.05),6组的IGF-1mRNA表达则显著高于1~5和7组(P<0.05)。粒细胞-巨噬细胞集落刺激因子(GM-CSF)与生长抑素(SS)融合表达质粒(pGM-CSF/SS)的GHR和IGF-1基因mRNA表达高于pGM-CSF+pcS/2SS共同免疫;同一DNA疫苗,口服免疫组GHR、IGF-1基因mRNA的表达总体上比肌肉注射组高。结果证明,GM-CSF可促进生长抑素DNA疫苗的免疫效果,提高肌肉和肝脏组织中GHR、IGF-1基因的mRNA表达;pGM-CSF/SS效果优于pGM-CSF+pcS/2SS,口服免疫组优于肌肉注射免疫组。
To determine the effect of pGM-CSF/SS on the mRNA expression of growth hormone receptor (GHR) and insulin-like growth factor-1 (IGF-1), seventy 22-day-old mice with half males and half females were separated into seven groups randomly, including control group (Group 1) and experimental group (Group 2-7). The mice in Group 2-4 were intramuscularly immunized with pcS/2SS, pGM-CSF/SS and pGM-CSF+pcS/2SS, respectively, and the animals in group 5- 7 were orally administered with attenuated Salmonella which harbored pcS/2SS, pGM-CSF/SS and pGM- CSF+pcS/2SS, respectively, while the control group was treated with normal saline. The expression level of GHR and IGF-1 mRNA in liver was detected by semi-quantitative RT-PCR, and the data showed that GHR mRNA expression lev- els of group 3, 5 and 6 were higher than those of group 1, 2, 4 and 7 (P〈0.05). IGF-1 mRNA expression level of group 6 was higher than those of group 1, 2, 3, 4, 5 and 7 (P〈0.05). GHR and IGF-1 mRNA expression levels of pGM- CSF/SS were higher than those of co-immunization of pGM-CSF+pcS/2SS. GHR and IGF-1 mRNA expression levels of the oral administration were largely higher than that of the muscular injection with the same DNA vaccines. These results suggest that GM-CSF can enhance immune effect of SS DNA vaccine and increase GHR and IGF-1 mRNA expression in liver tissue of mice. The immune effect of pGM-CSF/SS was superior to that of pGM-CSF+ pcS/2SS. The immune response of oral administration was higher than that of the muscular injection.
