“肺肠同治”对急性肺损伤大鼠肺和大肠组织P38表达影响 被引量:4

Effect of "Treating the Lung and the Intestine Simultaneously" on the Expression of P38 in Lung and Large Intestine in ALI Rats

摘要 [目的]探讨中医治肺、治肠及肺肠同治三法对脂多糖(Lipopolysaccharide,LPS)诱导急性肺损伤(Acute lung injury,ALI)大鼠肺和大肠组织中P38磷酸化的作用。[方法]SD大鼠72只随机分为6组,即对照组、模型组、芪冬活血饮组、大黄汤组、地塞米松组(DXM组)和芪冬活血饮加大黄组,采用气道内滴入LPS制备ALI模型,造模前24h、12h及造模后12h分别采用相应药物干预,造模后24h处死大鼠,采集标本,Western-blot检测肺及大肠P38的表达情况及观察肺组织病理损害。[结果]P38的表达在模型组最高,NS组最低,同其它各组比较差异具有显著性(P<0.05),芪冬活血饮及大黄汤可不同程度抑制肺、大肠组织P38磷酸化,两者合用时具有协同作用,对P38抑制作用更加明显,疗效同地塞米松组相当。[结论]肺肠同治能抑制LPS诱导的ALI大鼠肺肠组织P38磷酸化,减轻肺组织病理损害,疗效具有协同性,优于单纯治肺或单纯治肠组。 [Objective]In order to investigate the expression of P38 phosphorylafion in lung and large intestine in ALl rats induced by LPS after intmgastric administration with Qidong Huoxue Yin(treating lung),Rhubarb(treating intestine),Qidong Huoxue Yin plus Rhubarb(treating the lung and the intestine simultaneously) respectively. [Method]72 SD rats were randomly divided into six groups:control group,model group,Qidong Huoxue Yin group,Rhubarb group, Dexamethasone group(DXM group) and Qidong Huoxue Yin plus Rhubarb group.Normal saline was dripped into trachea direcdy in control group while other groups with LPS.The intragastric administration was performed in each group with the corresponding drug before and after LPS mold- building.Specimens were obtained 24h after LPS mold-building.The expression of P38 in lung and large intestine was detected by western-blot,and HE staining was used to examine lung pathological destruction.[Result]The expression of P38 was the lowest in control group and highest in model group, and had a statistically significant difference(P〈0.05) compared to other groups. Both Qidong Huoxue fin and Rhubarb could inhibit P38 phosphorylation ex- perssion in lung and large intestine.Besides, a synergetic effect was observed when Qidong Huoxue Yin was used with rhubarb,which had no statistically difference compared with dexamethasone group.[Conclusion]The "treating the lung and the intestine simultaneously" treatment can inhibit P38 phosphory- lation expression, attenuate lung pathological destruction, and it's more effective than treating lung or large intestine respectively.
