毛细管电泳pH酸修饰在线富集方法检测唾液中鸦片类毒品 被引量:2

摘要 建立了唾液中鸦片类毒品吗啡、可待因和6-单乙酰吗啡的毛细管电泳pH酸修饰在线富集方法,并进行了方法学考察。100mmol/L的磷酸盐缓冲液(含20%甲醇和5%异丙醇)作为背景缓冲液,10kV下电迁移进样99s,然后电迁移(2kV,2s)引入一段100mmol/L的盐酸区带,最后施加25kV电压进行分离检测。3种毒品在20~1 000ng/mL范围内具有良好的线性关系,r为0.9934~0.9998,检出限均为7ng/mL,相对标准偏差(RSD)<19%(n=5)。该方法样品前处理简单,只需用甲醇稀释样品便可进样分析,方法灵敏度较高、稳定性较好,可用于生物检材中痕量鸦片类毒品的检验。 Acid-mediated stacking in capillary electrophoresis (CE) was developed and validated for opiates analysis in saliva in the present paper. Optimum conditions for the determination of morphine, codeine and 6-monoacetyl morphine in acid-mediated CE were investigated. 100 mmol/L phosphate buffer (containing 20% methanol and 5% isopropanol,pH=3) was used as the background electrolyte for CE separation. The samples were introduced electrokinetically for 99 s at 10 kV in the stacking. Acid plug (100 mmol/L HC1) was introduced for 2 s at 2 kV after sampling. The methods showed a significant increase in LODs(7 ng/mL),while providing good separation efficiency and reproducibility(RSD≤19 %, n=5). Besides, the sample pretreatment was simple(only diluted with methanol). The method can be used to analyze trace amount of opiates in biological samples.
