摘要
目的 :检测Fas基因表达并观察转染Fas基因对卵巢癌细胞凋亡的影响 ,探讨其在卵巢癌发生、发展中的作用。方法 :经流式细胞术、RT PCR方法检测 6种卵巢癌细胞Fas的表达水平。构建pcDNA3 Fas真核表达载体 ,经脂质体转染细胞 ,通过流式细胞仪检测Fas基因表达变化。应用PI染色经流式细胞术检测观察转染Fas基因对3AO细胞凋亡的影响。结果 :6种卵巢癌细胞均表达Fas,均属中低表达。经 pcDNA3 Fas真核表达载体转染的卵巢癌细胞Fas基因表达及对Fas单抗诱导凋亡的敏感性均有不同程度提高。结论 :Fas基因低表达及对Fas介导凋亡的抵抗可能与卵巢癌发生、发展有关 ;转染Fas基因可提高Fas基因表达 。
Objective: To study the expression of Fas gene and investigate apoptosis in ovarian carcinoma cells transfected by Fas, so as to explore the role of Fas gene in the development and progression of ovarian carcinoma. Methods: The expression of Fas gene in 6 strains of ovarian carcinoma cells was detected with flow cytometry and RT PCR methods. The eukaryotic expression vector pcDNA3 Fas was constructed and transfected into ovarian carcinoma cells with lipofectin, and then the change in expression of Fas gene was determined by flow cytometry. By means of PI staining flow cytometry the effect of this gene on apoptosis of transfected 3AO cells were observed. Results: Fas gene was expressed in 6 ovarian carcinoma cells, but the proportion of Fas expression was low. Transfection of ovarian carcinoma cells with the eukaryotic vector pcDNA3 Fas enhanced cell surface expression of Fas gene and induced susceptibility to Fas antibody mediated apoptosis. Conclusion: Down regulation of Fas expression and resistance to Fas mediated apoptosis in ovarian carcinoma cells were found to be related to the development and progression of ovarian carcinoma cells. Fas gene transfer may increase Fas expression in ovarian carcinoma cells and enhance their apoptosis.
