摘要
目的 :研究六亚甲基双乙酰胺 (HMBA)对涎腺粘液表皮样癌高转移细胞 Mc3恶性表型的影响。方法 :采用 MTT法、细胞计数法、克隆形成法、流式细胞术及癌细胞裸鼠尾静脉注射法 (每只鼠 10 6个细胞 )研究HMBA对 Mc3细胞生长以及转移力的抑制作用。结果 :HMBA在 1mmol/ L 时对 Mc3细胞生长抑制率为36 % ,作用于 Mc3细胞 5 d后对照组和处理组细胞倍增时间 (h)分别为 2 0 .8和 2 3.5 ,S细胞比例 (% )为 2 4.5和2 3.0。野生型 P5 3蛋白表达率 (% )为 2 4.0和 99.7,nm2 3- H1 蛋白表达率 (% )为 99.9和 98.9,克隆形成率 (% )为 2 3.5和 16 .0 ,在裸鼠肺表面转移结节数为 139± 6 1和 83± 37。结论 :HMBA可抑制 Mc3细胞的转移力 。
Objective: To study the effects of hexamethylene bisacetamide(HMBA) on the metastatic potential of mucoepidermoid carcinoma Mc3 cells. Methods: The effects of HMBA on the metastatic potential of Mc3 cells were studied with MTT assay, cell counting, flow cytometry and tail vein injection of the cells into nude mice(10 6 for each).Results: At the dose of 0.001 6~1 mmol/L HMBA inhibited Mc3 cell growth by 6.7%~36.1% in a dose dependant way. After the cells had been treated with 1 mmol/L of HMBA for 5 d the doubling time of the cells was 23.5 h, the percentage of S phase cell 23.0%, that of wild type P53 protein expression 99.7%, nm23 H 1 98.9%, clonogenecity 16.0%, number of metastatic foci on lung surface 83±37, respectively; while those of untreated cells were 20.8 h, 24.5%, 24.0%,99.9%,23.5% and 139±61 respectively.Conclusion: HMBA may inhibit the metastatic potential of Mc3 cells and the effects may be related to differentiation induction.
