猕猴耳廓软骨细胞体外不同的培养方式与生长活性 被引量:5

摘要 目的 :探讨猕猴耳廓软骨细胞在体外不同介质的培养情况及其生长活性。材料与方法 :对 6只猕猴进行耳廓软骨取材、软骨细胞的分离 ,并行单层贴壁培养及在人工基质上的培养。通过倒置显微境和扫描电镜观察不同方式培养的细胞生长情况。结果 :单层贴壁培养的软骨细胞可传代 5~ 6代 ,但第三代软骨细胞分泌的Ⅰ型与Ⅱ型胶原的能力无明显差别 ;在几丁质上培养的软骨细胞能保持体内细胞的正常形态 ;在VICRYL上培养的软骨细胞能分泌软骨基质。结论 :不同的体外培养方式下软骨细胞的生长活性略有不同 ,需根据不同的实验要求选择相应的培养方式。 Objective:To observe the cellular activities with different cultivations of chondrocytes of auricular cartilages from rhesus monkeys.Methods:The chondrocytes of auricular cartilages from 6 rhesus monkeys were isolated and cultured in monolayer and in matrices.The changes of cellular morphology were investigated with inverted microscope and scanning electron microscope(SEM).The cellular activities were studied with immunohistochemistry(IHC).Results:The chondrocytes cultured in monolayer were polygonal,and IHC showed that there was no significant difference between type Ⅰ and type Ⅱ collagen stain in 3rd generation.The morphology of chondrocytes cultured in chitin were spherical,similar with the morphology in vivo.The chondrocytes cultured in VICRYL can secret ground substance of cartilage.Conclusions:The cellular activities with different cultivations of chondrocytes is somehow different.
