摘要
采用海藻酸钠包埋法制备固定化漆酶,结果表明,固定化pH值为4.5,海藻酸钠与CaCl2质量浓度均为2%,酶液与海藻酸钠体积比为1∶1时,漆酶的固定化效果最佳,酶活回收率为48.6%。固定化酶的最适pH值为3.0,最适温度为40℃,米氏常数K m为21.5μmol/L,游离酶的最适pH值为3.0,最适温度为30℃,米氏常数K m为18.6μmol/L,与游离酶相比,固定化漆酶的热稳定性有明显提高,耐酸性略有增加,最适反应温度提高了10℃。
With the encapsulation of calcium-alginate beads, laccase was immobilized. The results showed that when the immobilized pH value was 4.5, the concentration of sodium alginate and CaC12 were both 2%, and the volume ratio between enzyme solution and sodium alginate was 1:1, lac- case immobilization got best results. The recovery of enzyme activity was 48.6%. The optimum pH value and optimum reaction temperature of im- mobilized enzyme were 3.0 and 40℃, Michaelis constant of immobilized enzyme was 21.5μmol/L. The optimum pH value and reaction temperature of free enzyme were 3.0 and 30℃, Michaelis constant of free enzyme was 18.6μmol/L. Compared with free enzyme, the thermal stability of immobi- lized enzyme had been significantly improved, the acid resistance increased slightly, and the optimum reaction temperature increased by 10℃.
