参麦注射液对脓毒症模型大鼠肾脏细胞凋亡和Bcl-2/Bax蛋白表达的影响 被引量:3

摘要 目的:观察参麦注射液对脓毒症模型大鼠肾脏细胞凋亡及其Bcl-2和Bax蛋白表达的影响。方法:45只SD大鼠随机分为假手术组、脓毒症模型组和参麦组,每组15只。采用盲肠结扎穿刺法(CLP)造模。脓毒症模型组术前1h腹腔内注射生理盐水10mL/kg,假手术组除不结扎和穿刺盲肠外,其余操作同脓毒症模型组。参麦组术前1h腹腔内注射参麦注射液10mL/kg。术后24h取大鼠肾脏组织,采用TUNEL法检测并计算肾脏细胞凋亡指数(AI),免疫组化检测Bcl-2和Bax蛋白表达。结果:三组大鼠体质量差异无统计学意义(P>0.05),三组大鼠均可见肾脏细胞凋亡,细胞凋亡多发生于肾小球细胞。脓毒症组和参麦组肾脏细胞AI均高于假手术组(P<0.01),参麦组肾脏细胞AI虽低于脓毒症组,但差异无统计学意义(P>0.05);与假手术组比较,脓毒症组和参麦组肾脏细胞Bcl-2表达下降(P<0.01,P<0.05),Bax表达上调(P<0.01),Bcl-2/Bax比值减小(P<0.01)。与脓毒症组比较,参麦组Bcl-2明显上调(P<0.01),Bax表达下调(P<0.05),Bcl-2/Bax比值增大(P<0.01)。结论:参麦注射液能上调脓毒症模型大鼠肾脏细胞Bcl-2蛋白表达,下调Bax蛋白表达,减少脓毒症大鼠肾脏细胞凋亡。 Objective:To investigate the effects of Shenmai injection on renal apoptosis and Bcl-2/Bax protein expres- sion in septic rats. Methods:Forty-five male SD rats were randomly divided into sham operation group,sepsis group,and drug group,15 rats in each group. The sepsis model was made by cecal ligation and puncture(CLP) modeling. In the sepsis group the rats were injected with 10 mL/kg saline into their peritoneal cavities 1 h before CLP. The rats in the sham operation group were treated the same way except no ligation and puncture of the ce- cum. In the drug group the rats were injected with 10 mL/kg Shenmai injection fluid into their peritoneal cavities 1 h before CLP. All rats were given subcutaneous injection of 50 mL/kg Ringer's solution for anti-shock immedi- ately after operation. In 24 h after operation, renal specimens of the rats were collected. The renal apoptosis was detected by using TUNEL method. The Bcl-2 and Bax protein expression was determined by using immunohisto- chemistry. Results:Six rats were died in the sepsis group and 4 rats in the drug group. No significant difference in body weight was noted among the three groups (P〉0.05). The renal apoptosis index in the sepsis and drug groups were higher than that in the sham operation group (P〈0.01),while that in the drug group was lower than that in the sham operation group without a significant difference (P=0.084). Compared with the sham operation group,the Bcl-2 protein decreased(P〈0.01 ,P〈0.05) and the Bax protein increased (P〈0.01),and the Bcl-2/Bax ratio reduced (P〈0.01) in the sepsis and drug groups. Compared with the sepsis group,the Bcl-2 protein increased(P〈0.01) and the Bax protein decreased (P〈0.05),and the Bcl-2/Bax ratio increased (P〈0.01) in the drug group. Conclusion: Shenmai injection can upregulate the Bcl-2 protein and downregulate the Bax protein,resulting in prevention from apoptosis in rats.
