摘要
[目的]研究反义寡核苷酸干涉家蚕非编码RNA表达的情况。[方法]构建Bm-15的反义寡核苷酸探针,通过脂质体转染昆虫Sf9细胞干涉Bm-15的表达。[结果]转染后细胞中Bm-15的表达量下降了63%,差异达显著水平,表明反义寡核苷酸探针对Bm-15基因表达有较好的抑制作用。[结论]该研究为进一步研究非编码RNA的功能奠定了良好的基础。
[ Objective] The aim was to study the interference of non-coding RNAs by antisense oligonucleotides (ASO) in silkworm. [ Meth-od] The ASO probe of Bin-15 was designed, and transfected to St9 cells by liposome to interference the expression of Bin-15. [ Result] The expression of Bm-15 decreased by 63% after ASO transfection, and the difference reached significant level, which showed that the ASO probe had good inhibitory effect on Bin-15 gene expression. [ Conelusion] The study laid a good foundation for the further study on the function of non-codina RNAs.
