Effects of thyroid hormone receptor on energy and lipid metabolism

摘要 甲状腺激素(TH)对能量与脂代谢具有重要的调节作用.近年来,随着甲状腺激素受体(TR)亚型的发现,众多研究者对其在代谢调节中的功能进行了一系列研究.TR在人体各组织广泛存在,其中TRβ与TH结合后,能降低胆固醇与甘油三酯水平.目前,TRβ激动剂已作为新型降脂药进入临床研究,它可能通过逆向胆固醇转运及非胆汁性胆固醇排泄的通路达到降低血脂的效果.未来TR在降脂及减肥方面的前景日益得到广泛关注,它极有可能成为众多治疗脂代谢紊乱及相关疾病的新靶点. Thyroid hormone (TH) plays a significant role in regulating both energy and lipid metabolism.Since the subtypes of thyroid hormone receptor (TR) have been explored and reported several years ago,numerous studies have been focused on their roles in metabolic regulation.TR can be extensively found in tissues of the human body.The binding of TH and TRβ can lower the levels of cholesterol and triglyceride.Currently,agonist of TRβ has been in clinical trials as a new lipid lowering agent,possibly via reverse cholesterol transport and trans-intestinal cholesterol excretion.The impacts of TR on lipid and energy metabolism are attracting increasingly intense attention.It is prospectively promising to fight clinically against hyperlipidaemia and obesity.It is most likely to grow to be a new target for lipid metabolism disorders and related diseases.
