游离大黄蒽醌对骨质疏松模型大鼠骨密度和骨矿含量的影响 被引量:5

Effects of Free Anthraquinone on Bone Mineral Density and Bone Mineral Content of Osteoporosis Model Rats

摘要 目的:研究游离大黄蒽醌类化合物对骨质疏松模型大鼠骨密度(BMD)、骨矿含量(BMC)的影响。方法:灌胃醋酸泼尼松(3.5 mg/kg),每日1次,以复制大鼠骨质疏松模型。4月龄126只雌性SD大鼠随机均分为正常对照(等容生理盐水)组、模型(等容生理盐水)组、游离大黄蒽醌(70 mg/kg)组,每组再分1、3、5、7、9、11、13周7个时间点。上午灌胃游离大黄蒽醌,下午灌胃醋酸泼尼松。在相应时间点,大鼠麻醉后心脏抽血至尽处死,采集四肢骨,以pQCT骨密度仪分别扫描大鼠股骨全骨和股骨远端(2 mm),测定BMD和BMC。结果:与正常对照组比较,复制模型1周后,模型组大鼠股骨远端BMD显著减小;复制模型3周后,模型组大鼠股骨全骨BMD显著减小;复制模型5周后,模型组大鼠股骨远端BMC显著减少(P<0.05)。与模型组比较,在7个时间点上游离大黄蒽醌组大鼠股骨全骨和股骨远端的BMD显著增加(P<0.05)。结论:游离大黄蒽醌可预防醋酸泼尼松所致的股骨BMD的丢失,未观察到大鼠股骨BMC随时间出现显著蓄积的现象。 OBJECTIVE: To investigate the effects of free anthraquinone compounds on bone mineral density(BMD) and bone mineral content(BMC) in osteoporosis model rats. METHODS: Osteoporosis model was induced by intragastrical administration of prednisone acetate (3.5 mg/kg) once a day. 126 female SD rats aged 4 months were randomly divided into normal control group (constant volume of normal saline), model group (constant volume of normal saline) and free anthraquinone group (70 mg/kg). Each group was further subdivided into 7 subgroups according administration period of 1,3,5,7,9, 11,13 weeks. They were given relevant medicine intragastrically in the morning and prednisone acetate intragastrically in the afternoon. At corresponding time points, the heart blood of rats was exhausted after anaesthesia and those rats were sacrificed, and bones of limbs were collected. The left femur bone (all) and femur distal part (2 ram) were scanned by peripheral quantitative computed tomography (pQCT) re- spectively to determine BMD and BMC. RESULTS: Compared with normal control group, the BMD was significantly decreased in model group at femur distal part 1 week after duplicating model. The BMD was significantly decreased in model group at femur bone(all) 3 week after duplicating model. The BMC were significantly decreased in model group at femur distal part 5 week after duplicating model. Compared with model group, the BMD were significantly increased in free anthraquinone group both at femur bone (all) and femur distal part at 7 time points(P〈0.05) ; the BMD of femur bone (all) were increased gradually at 1-7 time points. CONCLUSIONS: Free rhubarb anthraquinone can prevent the prednison-induced bone loss, but there is no BMC accumula- tion in the femur bone of rats as time.
