全氟化碳对大鼠心肌缺血再灌注损伤的预防作用 被引量:1

Protective effects of perfluorocarbons on myocardial ischemial and reperfusion injury in rat

摘要 目的:观察全氟化碳对大鼠心肌缺血再灌注损伤的预防作用。方法:将45只雄性Wistar大鼠随机分为假手术组、缺血再灌注组与全氟化碳组。全氟化碳组在缺血前48h每天腹腔注射全氟化碳(30ml/kg)。结扎大鼠冠状动脉左前降支,使心肌缺血30min,再灌注120min,制成心肌缺血再灌注损伤模型。分别测定血清中乳酸脱氢酶(LDH)、超氧化物岐化酶(SOD)、丙二醛(MDA)、髓过氧化物酶(MPO)水平,并行HE染色观察缺血心肌的病理变化。结果:与假手术组比较,缺血再灌注组LDH、MDA、MPO值均增高,而SOD活性降低(P<0.05);与缺血再灌注组比较,全氟化碳组LDH、MDA、MPO值均降低,而SOD活性增高(P<0.05),且心肌形态学改变更小。结论:全氟化碳对心肌缺血再灌注损伤有预防作用,可能与抗氧化和抗炎症反应有关。 Objective:To observe the protective effects of perfluorocarbons on myocardial ischemial and reper- fusion injury in rats. Method: The 45 male wistar rats were divided randomly into sham operation group, myocardi- al ischemia and reperfused injury (IRI) group and perfluorocarbon (PFC) group. Perfluorocarbons (30 ml/kg) were intraperitoneally injected 48 hours before ischemia everyday. The left anterior descending branch of coronary artery was ligated for 30 minutes and reperfused for 2 hours to build the myocardial ischemial and reperfusion inju- ry model. The changes of lactate dehydrogenase (LDH), superoxide dismutase (SOD), malondialdehyde (MDA), myeloperoxidase (MPO) levels were determined, and the pathological changes of myocardium were observed by HE stain. Result: Compared with sham operation group, LDH, MDA and MPO increased and SOD decreased in IRI group (P^0.05). Compared with IRI group, LDH, MDA and MPO decreased and SOD inecreased in PFC group (P^0.05), and PFC inhibited the histomorphological changes of ischemic myocardium. Conclusion:perfluorocarbon relieves the ischemia and reperfusion injury and the cardioprotection may be due to antioxidant effects and antiinflammatory reaction.
