三七预防糖尿病大鼠视网膜微血管病变的实验研究 被引量:8

摘要 目的探讨早期口服三七对糖尿病大鼠视网膜微血管病变的影响。方法 60只SD大鼠随机平均分成4组,A:正常大鼠生理盐水组;B:正常大鼠三七组;C:糖尿病大鼠生理盐水组;D:糖尿病大鼠三七组。C、D组尾静脉注射20 g·L-1STZ(55 mg·kg-1)制作糖尿病模型成功后三七治疗组每天按400 mg·kg-1三七粉灌胃给药一次,生理盐水组每天给予等量的生理盐水灌胃,12周后检测各组大鼠血液流变学、视网膜血管渗透性和毛细血管周细胞数目等指标。结果 A组、B组各项指标差异无统计学意义(P>0.05);与A组和B组相比,C组和D组大鼠出现明显的血小板黏附率、聚集率升高、视网膜血管渗透性增加、毛细血管周细胞数目减少(P<0.01);D组较C组糖尿病大鼠的视网膜渗透量降低了26.13%,差异有显著统计学意义(P<0.01)。D组糖尿病大鼠较C组糖尿病大鼠的周细胞计数降低了24.14%,但差异无统计学意义(P>0.05)。经三七治疗的D组糖尿病大鼠血小板黏附率(33.02±2.10)%、血小板聚集率(74.83±3.71)%,与C组(40.50±3.62)%和(84.00±6.48)%相比明显下降(P<0.05)。结论三七可降低糖尿病大鼠血黏度,减少视网膜血管渗透性,保护毛细血管结构,对早期糖尿病大鼠视网膜微血管病变具有一定的防治作用。 Objective To investigate the influence of oral Panax notogiseng ondiabetic retinopathy in rats at the early stage. Methods Sixty SD rats were randomly and evenly divided into 4 groups:A:normal rats with normal saline;B:normal rats with Panax notogiseng; C : diabetic rats with normal saline; D: diabetic rats with Panax noto- giseng. When diabetic model was successfully established by tail intravenous injection of 20 g · L-1 STZ (55 mg · kg- 1 ) to group C and group D, Panax notogiseng powder was administrated intragastrically to Panax treatment group one time each day with a dosage of 400 mg ~ kg - ~, the normal saline group was given the same dosage of normal saline in the same manner. Hemorheology,retinal vascular permeability and the number of capil- lary pericytes were tested on all rats 12 weeks later. Results No statistical signifi- cance was found in all indicators of group A and group B (P 〉 0.05 ) ; The platelet adhe- sion rate and aggregation rate significantly increased, retinal vascular permeability raised and the number of capillary pericytes reduced (P 〈 0.01 ) in group C and D, compared with group A and B;Retinal vascular permeability in group D lowered 26.13% than that in group C,and the difference had significant statistical significance(P 〈0.01 ). Count of capillary pericytes in group D reduced by 24.14% than that in group C,but no statistical significance was found ( P 〉 0.05 ). The platelet adhesion rate ( 33.02 ± 2. l0 ) % and platelet aggregation rate (74.83 ± 3.71 )% in group D were decreased markedly than those in group C,which were (40.50 ±3.62)% and (84.00±6.48)% ,respectively (P 〈 0.05 ). Conclusion Panax notogiseng may reduce blood viscosity and retinal vascular permeability as well as protect the blood capillary structure in diabetic rats. It also may prevent the diabetic retinopathy at an early stage.
