摘要
目的:建立多波长高效液相色谱法,同时测定四神丸中五味子醇甲、五味子甲素、五味子乙素、补骨脂素、异补骨脂素、去氢二异丁香酚、吴茱萸次碱7个有效成分的含量。方法:采用Diamond C18色谱柱(4.6 mm×250 mm,5μm),柱温30℃,以乙腈-0.1%磷酸水溶液为流动相梯度洗脱(0 min,30%A;10 min,40%A;30 min,50%A;60 min,90%A;61~70 min,30%A),检测波长为220 nm(五味子醇甲、五味子甲素、五味子乙素)、280 nm(补骨脂素、异补骨脂素、去氢二异丁香酚)和345 nm(吴茱萸次碱)。结果:7个有效成分达到完全分离;线性关系良好;加样回收率在94.8%~104.1%。结论:建立的方法准确可靠,可用于四神丸的质量控制。
Objective:To develop an HPLC method with multiple wavelength ultraviolet detection for the simultane- ous determination of seven active components ( schisandrin, deoxyschizandrin,γ schisandrin, psoralen, isopsor- alen, dehydrodiisoeugenol, rutaecarpine) in Sishen pills. Methods:The samples were separated on a Diamond C18 column (4.6 mm × 250 mm, 5μm) by gradient elution (0 min, 30% A ; 10 min, 40% A ; 30 min, 50% A ; 60 min, 90% A ;61 - 70 min,30% A) using acetonitrile - 0. 1% phosphoric acid solution. The eluate was detected at multi- ple wavelengths of 220 nm for schisandrin, deoxyschizandrin and γ- schisandrin, 280 nm for psoralen, isopsor- alen, dehydrodiisoeugenol and 345 nm for rutaecarpine. Results:The complete separation was obtained for the sev- en active compounds. The seven regression equations showed good linear relationships. The average recoveries of the compounds above were between 94. 8% and 104. 1%. Conclusion:The method is feasible, accurate and sta- ble, which can control the quality of Sishen pills.
