摘要 目的探讨活化白细胞黏附分子(ALCAM)在胃癌组织中的表达水平并预测其上游调控基因。方法采用SYBR GreenⅠ实时荧光定量PCR及免疫组织化学方法分别检测27对和50对胃癌及癌旁正常组织中ALCAM的mRNA和蛋白表达水平,并分析其表达情况与临床病理资料的相关性。用生物信息学软件预测ALCAM的调控基因。结果胃癌组织中ALCAM的mRNA和胞膜蛋白表达水平高于癌旁正常组织(P<0.05),且与淋巴结转移有关(P<0.05),但与患者的年龄、性别、肿瘤组织学分级及TNM分期无关(P>0.05)。用生物信息学软件预测ALCAM可能为miR-9、miR-192、miR-215的靶基因。结论 ALCAM的mRNA及胞膜蛋白在胃癌组织中高表达,且与淋巴结转移有关。ALCAM在胃癌的高表达可能与微小RNA调控异常相关。 Objective To investigate the expression of activated leukocyte cell adhesion molecules (ALCAM) in gastric carcinoma and to predict the upstream regulatory factors of ALCAM. Methods ALCAM mRNA and protein expression levels in both gastric carcinoma and its adjacent non - tumor tissues were measured by SYBR Green I real - time quantitative PCR and immunohistochemistry. The bioinformatics software was applied to predict the regulatory factors of ALCAM. Results The mRNA and membranous expression of ALCAM in gastric carcinoma were significantly up - regulated than that in the adjacent normal tissues ( P 〈 0. 05 ), which was significantly correlated with lymph node metastasis ( P 〈0.05), while no significant correlation was revealed with age, gender, tumor histologic grade or TNM stage (P 〉 0. 05 ). According to bioinformaties software prediction, ALCAM was probably the target gene of miR -9, miR - 192, and miR - 125 gene. Conclusion The mRNA and membranous expression of ALCAM are up - regulated in gastric carcinoma tissue, and correlated with lymph node metastasis. Overexpression of ALCAM is probably due to the aberrant microRNA regulation.
