不同前处理方法测定小飞蓬植株中莽草酸含量比较

摘要 施用草甘膦后杂草体内莽草酸的含量可作为检测杂草对草甘膦抗性水平的重要指标,但样品的前处理方法对测定结果具有一定的影响。以小飞蓬(Conyza Canadensis L.Crong)为材料,设计了液氮速冻干燥研磨、常温匀浆和高温(120℃)杀青后真空干燥研磨3种样品前处理方法处理小飞蓬样品,用盐酸提取莽草酸,经高碘酸、氢氧化钠、甘氨酸显色,在380nm下测定光密度。3种前处理方法平行30次测定莽草酸含量平均值分别为563.3507、462.8153、888.1420μg/g,在5%水平上,3种处理具有显著差异。各处理标准误的大小分别为13.7343、23.7483、30.1076。液氮速冻干燥研磨前处理方法测定结果标准误小,准确度高,可作为莽草酸法检测杂草对草甘膦抗性水平中推荐的样品前处理方法。 The shikimate content in weeds treated by glyphosate was an important indicator to detect the glyphosate-resistance level of weeds,but the analysis results were usually influenced by the different pretreatment methods.In the trial,the horseweed was studied as experimental material,pestled after frozen in liquid nitrogen,homogenized at room temperature and pestled after vacuum drying with de-enzyme at high temperature(120℃)were designed to treat horseweed,and shikimate was extracted by hydrochloric acid,colored by periodic acid,sodium hydroxide and glycine.The absorbance was determined under 380nm,finally.The shikimate content averages of 30 repeats were 563.3507,462.8153μg/g and 888.1420μg/g respectively,with a significant difference at the 5% level.The standard errors of three treatments were 13.7343, 23.7483 and 30.1076 respectively.Pestled after frozen in liquid nitrogen treatment was a recommended pretreatment method in shikimate content method to detect the glypho-sate-resistant levels of weeds,because the lowest standard error and highest accuracy.
