交配与未交配三疣梭子蟹肝胰腺蛋白质组学的初步分析 被引量:1

摘要 采用双向电泳和基质辅助激光解析串联飞行时间质谱(MALDI-TOF/TOF)研究三疣梭子蟹交配与未交配雌性个体肝胰腺蛋白质表达.使用PDQuest软件分析双向电泳凝胶图像,结果显示:交配组和未交配组凝胶图像上分别能检测到(400±25)和(530±16)个蛋白点,其中共有15个蛋白点表达量有显著性差异(P<0.05).与未交配组相比,交配组三疣梭子蟹肝胰腺中6个蛋白点的表达量显著性上升.经质谱鉴定为:磷酸丙糖异构酶、磷酸丙酮酸水合酶、过氧化氢酶、δ-谷胱甘肽S-转移酶、ATP合酶α-亚基和胰凝乳蛋白酶.此外,交配组三疣梭子蟹肝胰腺中9个蛋白点的表达量显著性下降.经质谱鉴定为:延伸因子2、长链酰基CoA脱氢酶、假血蓝蛋白1、假血蓝蛋白2、异柠檬酸脱氢酶、线粒体乙醛脱氢酶X、转酮醇酶、血蓝蛋白α-亚基和腺苷高半胱氨酸酶A.该结果可为揭示三疣梭子蟹卵巢发育机理提供重要参考. Two-dimensional electrophoresis and matrix-assisted laser desorption ionization tandem time of flight mass spectrometry(MALDI-TOF/TOF) are used to analyze proteomics of hepatopancreas in mated and unmated female Portunus trituberculatus.The two-dimensional electrophoresis gel images analyzed using PDQuest software demonstrate that 15 spots are significantly distinctive.The quantitative expression of 6 spots is incremented in hepatopancreas in the mated group which is identified as triosephosphate isomerase,phosphopyruvate hydratase,catalase,δ-glutathione S-transferase,ATP synthase alpha subunit and chymotrypsin.In contrast,the quantitative expression of 9 spots is decremented in the mated group which is identified as elongation factor 2,long-chain acetyl-CoA dehydrogenase,cryptocyanin 1,cryptocyanin 2,isocitrate dehydrogenase,aldehyde dehydrogenase X mitochondrial-like,transketolase,hemocyanin alpha-subunit and adenosyl-homocysteinase A.The results provide important theoretical support for completing ovary maturation research on the female Portunus trituberculatus.
