摘要
采用鉴别液与Bouin's固定剥膜相结合方法,对虹鳟2细胞期、4细胞期、8细胞期、桑葚期、高囊胚期、原肠中期六个发育阶段,进行受精率计算方法研究。结果表明,计算鱼卵受精率取样时间从卵裂期开始,桑葚期最佳;取样数量随机抽取60粒,重复两次,求其平均值即可;鉴别液与Bouin's液固定剥膜后测定受精率差异不显著(P>0.05),生产中完全可以直接使用鉴别液来测定受精率,简单、快速、准确度高。此结果可以作为产沉性大卵虹鳟等冷水性鱼类受精率计算方法标准化依据。
The experiment performed computing method research of fertilization rate on six development states of rainbow trout that contains two cell stage, four cell stage, eight cell stage, morula, high blastula stage and gastrula metaphase by using two methods of differentiating fluid and Bouin's fluid. Evidence indicates that: sampling time of calculating fertilization rate should begin from ovum-cleavage stage and the best time is morula; Sampling quantity randomly should be drawn off 60 grain that was repeated two times and calculates mean value; fertilization rate that were be calculated by using differential fluid and Bouin's liquid were no significant differences(P〉0.05). The method is simply, rapid, highly accurate and completely could be directly applied in production. This result can be used the standardized basis of calculation method of fertilization rate as cold water fishes that produce sinkable eggs such as rainbow trout.
