摘要
目的探讨和建立多烯磷脂酰胆碱及其有关物质的定量分析方法。方法对比紫外与蒸发光散射检测器的灵敏度,比较流动相的溶剂组成对多烯磷脂酰胆碱及其有关物质色谱分离行为的影响,采用上述指标进行综合评价。结果采用蒸发光散射检测器;流动相A为正己烷-异丙醇-冰乙酸-三乙胺(20∶80∶0.35∶0.02),流动相B为甲醇-水-冰乙酸-三乙胺(80∶20∶0.45∶0.05),流速1 mL·min-1,梯度洗脱。结论所用色谱分析方法的系统适用性好、专属性强、灵敏度较高,适用于多烯磷脂酰胆碱及其有关物质的检测。
OBJECTIVE To explore and establish an available method for analysis of polyene phosphatidylcholine (PPC) and its related substances. METHODS Sensitivity of UV detector and evaporation light scattering detector (ELSD) and the effect of solvent composition in mobile phases on chromatographic separation behavior of PPC and its related substances were compared, and comprehensive evaluation of above indicators was carried out to optimize the analysis method. RESULTS The HPLC method was developed by using the following operating coalition : ELSD, mobile phase A n - hexane - isopropanol - acetic acid - triethylamine (20 : 80 : O. 35 : 0.02) and mobile phase B methanol - water - glacial acetic acid - triethylamine ( 80 : 20 : 0.45 : 0.05 ), a flow rate of 1 mL· min-1 ,gradient elution. CONCLUSION The method developed in this paper shows good specificity and high sensitivity, which is appropriate for detection of PPC and its related substances.
