补中益气汤不同配伍对脾虚发热大鼠模型胃肠功能的影响 被引量:14

摘要 目的比较补中益气汤不同配伍对脾虚发热大鼠模型胃肠功能的影响。方法雄性Wistar大鼠,200~220 g,随机分为正常对照组、脾虚发热模型组、补中益气汤原方组、补中益气汤去柴胡升麻组、补中益气汤大剂柴胡升麻组、单纯大剂柴胡升麻组共6组,每组10只。采用饮食失节+过度疲劳+注射脂多糖(LPS)法复制脾虚发热大鼠模型。每周测体重1次,并观察记录其外观行为变化。于第18天各给药组给予相应的中药灌胃,连续5天。第22天断头取血检测胃动素(GAS)、胃泌素(MTL)、生长抑素(SS)。结果与正常对照组比较,脾虚发热模型组大鼠的外观行为积分值均升高,体重明显降低,外周血GAS、MTL和尿D-木糖排泄率均明显降低,血SS含量升高。与脾虚发热模型组比较,补中益气汤原方组、补中益气汤去柴胡升麻组、补中益气汤大剂柴胡升麻组大鼠的外观行为积分值均降低,体重明显升高,外周血GAS、MTL和尿D-木糖排泄率均明显升高,血SS下降;单纯大剂柴胡升麻组的大鼠外观行为各指标积分值升高,体重降低,外周血GAS、MTL和尿D-木糖排泄率降低,血SS含量升高,差异均有显著或非常显著性意义(P<0.05或P<0.01)。结论脾虚发热大鼠存在明显的胃肠功能紊乱;补中益气汤能明显改善脾虚发热大鼠的胃肠功能,该方去升麻、柴胡或配伍大剂量升麻、柴胡,其改善作用均明显减弱,单纯大剂量升麻与柴胡配伍无改善作用,补中益气汤的健脾益气作用与方中升麻、柴胡及其剂量的选配有关。 Objective To compare the influences of Buzhong Yiqi Tang (BYT) with different compatibilities on gastrointestinal function in rats with spleen deficiency fever. Methods Male Wistar rats (weighted from 200 g to 220 g) were randomly divided into normal group, model group, BYT group, BYT without Chai-Sheng group, BYT with high-dose Chai-Sheng group and high-dose Chai-Sheng group (each n = 10). The rat model with spleen deficiency fever was established by using the methods of improper diet, over strain and injection of lipopolysaccharide (LPS). The body weight of rats in all groups was detected every week, and the changes of behavior were observed and recorded. On the 18th day all the groups were intragastrically given corresponding decoctions respectively for 5 consecutive days. On the 22th day the levels of gastrin ( GAS), motilin (MTL) and somatostatin (SS) in blood were detected in rats of all groups. Results In model group, the integral of behavior and level of SS in peripheral blood increased, body weight, levels of GAS and MTL in peripheral blood and D-xylose excretion rate in urine decreased significantly compared with normal group. In BYT group, BYT without Chai-Sheng group and BYT with high-dose Chai-Sheng group, the integral of behavior and level of SS in peripheral blood decreased, body weight, levels of GAS and MTL in peripheral blood and D-xylose excretion rate in urine increased significantly compared with model group. In high-dose Chai-Sheng group, the integral of behavior and level of SS in peripheral blood increased, body weight, levels of GAS and MTL in peripheral blood and D-xylose excretion rate in urine decreased ( P 〈 0.05 or P 〈 0.01 ). Conclusion Conspicuous gastrointestinal dysfunction appears in the rats with spleen deficiency with fever, which can be alleviated significantly by BYT. BYT without Chai-Sheng and BYT with high-dose Chai-Sheng have decreased curative effects, and the simple compatibility of high-dose Chai-Sheng has no curative effect. The effects of invigorating spleen and supplementing qi of BYT may be related to the dose and combination of Shengma ( Rhizoma Cimicifugae) and Chaihu ( Radix Bupleuri).
