肿瘤坏死因子-α对大鼠颈动脉体球细胞胞内钙的影响

The effect of tumor necrosis factor-α on the [Ca^(2+)]_i level in the glomus cells of the rat carotid body

摘要 目的:研究肿瘤坏死因子Ⅰ型受体(tumor necrosis factor receptor type Ⅰ,TNFR-Ⅰ)在大鼠颈动脉体(carotid body,CB)中的表达,以及TNF-α对CB球细胞胞内钙离子浓度([Ca2+]i)的影响。方法:Western Blot和免疫荧光双重染色检测TNFR-Ⅰ在大鼠CB中的表达,用钙成像技术检测TNF-α对大鼠CB球细胞[Ca2+]i的影响。结果:Western Blot结果显示,TNFR-Ⅰ阳性条带出现在55kD处,与其分子量一致。免疫荧光双重染色结果显示,TNFR-Ⅰ强烈表达在大鼠CB球细胞中。钙成像结果显示,外源性给予TNF-α可引起球细胞[Ca2+]i迅速升高。结论:大鼠CB球细胞表达TNFR-Ⅰ,可以感受促炎性细胞因子TNF-α的刺激。 Objective: To study the expression of tumor necrosis factor receptor type I (TNFR-I) in the rat carotid body ( CB), and the effect of TNF-α on intraeellular Ca2 + ( [ Ca2 +] i ) level in the glomus cells of the rat CB. Methods : West- ern Blot and double immunofluorescent double staining methods were used to investigate the expression of TNFR-I in the rat CB. Calcium imaging was used to observe the effect of TNF-oL on [ Ca2 +]i level in the glomus cells of the rat CB. Re- sults: The result of Western Blot shwed that TNFR-I protein band appeared at 55 kD, consistent with the molecular weight of the receptor. The result of double immunofluorescent staining showed that TNFR-I was expressed in the CB glo- mus cells. The result of calcium imaging showed that extracellular application of TNF-α induced a rise in [ Ca2+ ] i level in the cultured glomus cells of the rat CB. Conclusion: TNFR-I is strongly expressed in the glomus cells of the rat CB, and the glomus cells can sense the stimulation of proinflammatory cytokine TNF-α.
