脂质过氧化反应在酒精及四氯化碳致大鼠肝纤维化发生中的作用 被引量:2

摘要 目的:观察脂质过氧化反应在酒精及四氯化碳(CCl4)致大鼠肝纤维化中的作用。方法:健康雄性Wistar大鼠40只,随机分为4组,实验持续4周。A组(空白对照组):生理盐水灌胃,每天10ml/kg。B组(模型组):白酒灌胃每天10ml/kg,同时腹腔内每次注射CCl40.25ml/kg,每周2次。C组(阳性药物干预组1):在B组基础上预先灌胃硫普罗宁每天100mg/kg。D组(阳性药物干预组2):在B组基础上预先灌胃茴三硫每天20mg/kg。第2周末、第4周末分别取血检测ALT、AST、ALP、GSH-Px、MDA等项目。实验结束处死动物,取肝组织行病理检验。结果:酒精及CCl4可引起大鼠肝功能损害、脂质过氧化反应加重,组织学出现肝纤维化表现。使用2种药物干预后脂质过氧化反应受到抑制,肝功能和肝脏纤维化改变均有减轻,各组间差异有统计学意义。结论:脂质过氧化在大鼠酒精及CCl4致大鼠肝纤维化发生中起重要作用,抑制该反应可以显著抑制肝纤维化的形成。 Objective:To observe the role of lipid peroxidation in the pathogenesis of liver fibrosis induced by alcohol and carbon tetrachloride in rats.Methods :Forty male Wistar rats were randomly assigned to one of the four groups and the experiment was continued for 4 weeks.Group A(placebo group) was given orally by saline at the dose of 10ml/kg/d.The other groups were perfused gastrically by 56% alcohol at the dose of 10ml/kg/d, and at the same time CCl4 was injected intraperitoneally at the dose of 0.25ml/kg,b.i.w.Group C(intervention group 1)was beforehand administened orally with thiopronin at the dose of 100mg/kg/d.Group D(intervention group 2)was beforehand perfused gastrically by anethohrithione at the dose of 20mg/kg/d.At the end of 2nd week and the 4th week ALT,AST,ALP,GSH-Px,MDA were examined.When the experiment was over,rats were sacrificed and the histopathologic alterations of liver were observed.Results:At the 2nd week and 4th week,group B were obviously different from group A,C,D.Group B showed significant heptic injury,including increased ALT,AST,ALP,MDA and decreased GSH-Px,cellular necrosis,inflammatory infiltration and liver fibrosis.Treatment with above two drugs could inhibit the lipid peroxidation and liver fibrosis was mitigated. Conclusion:lipid peroxidation may play an important role in the pathogenesis of liver fibrosis induced by alcohol and carbon tetrachloride.Inhibition of lipid peroxidation may be an importment measure for alleviating liver fibrosis.
