摘要
采用正交优化试验,以可溶性总糖含量作为考察指标,对影响蒽酮比色法测定马铃薯淀粉可溶性总糖的测定条件的因素进行了研究。结果表明,马铃薯淀粉可溶性总糖的最佳测定条件为:可溶性总糖经80%乙醇提取10 min后,反应时间15 min,显色剂用量9 mL,反应静置时间30 min,反应温度95℃。在最佳测定条件下,马铃薯淀粉中可溶性总糖的含量为23.01 mg/100 g。
The factors of determination conditions of total sugar from potato starch by anthrone colorimetry was studied by the orthogonal test in this paper.The results showed that the optimum determination conditions were as follows : after extract soluble total sugar from potato starch for 10 min with 80 % ethanol, reaction time for 15 min, colour-display reagent consumption for 9 mL, reaction stalling time for 30 min,reaction temperature for 95 ~C.The yield can reach 23.01 mg/lO0 g under the optimum determination conditions.
