五指山小型猪近交系心、肝和肾的微观形态学研究

摘要 为了研究五指山小型猪近交系心、肝和肾的微观形态学特点。本试验选用2、4和12月龄五指山小型猪近交系18只(n=6),分别采用光镜和透射电镜技术对心、肝和肾的显微和超微结构进行观察。结果显示,随着月龄增加,五指山小型猪近交系心室壁的心内膜层逐渐增厚,心肌细胞排列逐渐密集,心肌细胞间出现大小不等的脂肪泡,心室肌细胞间闰盘结构不明显;12月龄心内膜内皮下层出现平滑肌样细胞团;肝小叶逐渐增大,小叶间结缔组织变薄,肝细胞排列密集,肝血窦内枯否氏细胞增多。肾小体数量增多,血管球体积增大,肾小囊变小,肾小管上皮细胞增厚。电镜下,12月龄五指山小型猪近交系心肌细胞胞质内肌原纤维明带和暗带清晰可见,其间夹有丰富的线粒体;肝细胞胞质内富含线粒体和游离的糖原颗粒,部分胞质内含有大小不一的脂滴,肝血窦面微绒毛不发达,狄氏隙内贮脂细胞较大,周围有丰富的网状纤维;肾小球毛细血管内皮细胞较厚,近端小管上皮细胞腔面有丰富的微绒毛,远端小管上皮细胞基底面质膜内褶不发达。以上观察结果表明,五指山小型猪心,肝和肾的微观形态结构与人和其它哺乳动物基本相似,但有其自身的特点。 In order to study the microscopic morphological characteristic of heart, liver and kidney in Wuzhishan minipig inbred (WZSP) , the microstructure and ultrastructure of 18 WZSPs (n = 6) with 2, 4 and 12 months old were observed by light microscopy and transmission electron microscopy techniques. The results showed that with increasing age, the thickness of endocardium in the ventricular wall increased, and the cardiomyocytes were densely arranged with fat cells vesicles of varying sizes among it, and the intercalated discs were not obvious in WZSP; the smooth muscle -like cells existed in the subendothelial layer of the endocardium in the 12 months old WZSP; the volume of hepatic lobule increased with densely arranged hepatocytes, the connective tissue were de- creased among the hepatic lobules, and kupffer cells were increased in the hepatic sinusoid ; the quantity of renal corpuscle were increased with larger glomerulus and smaller renal capsule, and the epithelial cells became thickin the renal tubule. In the electron microscope, the light and dark band of myofibril were clearly in the cardio- myocyte of 12 months old WZSP, and the abundant mitochondria existed among the cardiomyocyte; there was a large number of mitochondria and free glycogen granules in the cytoplasmic of hepatocyte with lipid droplets of varying sizes, the microvillies were not developed in the sinusoidal side of hepatocyte, the fat - storing cells were larger with abundant reticular fibers around it; the endothelial cells were thick in the capillaries of glomerular, the free surface of endothelial cells had abundant microvillies in the proximal tubule, the plasma membrane in- foldings were not developed on the base surface of endothelial cells in the distal tubule. Above results indicated that the microscopic morphology of the heart, liver and kidney in WZSP were similar to that of human and other mammal, but had itself characteristic.
