落花生属植物基因组DNA提取及鉴定

摘要 以落花生(ArachisL.)嫩叶为材料,在传统CTAB法的基础上加以改良,建立了落花生基因组DNA的CTAB提取方法。结果表明:所提取的DNA经核酸蛋白含量测定得A260/A280处于1.84~1.95,经琼脂糖凝胶电泳检测各样品均具有一条明亮主带且无拖尾现象。表明改进的CTAB法提取得到的DNA完整性好,纯度高,可有效应用于后续的分子标记等研究。 The extraction of genomic DNA from Arachis L. is a precondition of molecular biological research. An improved method of extracting genomic DNA from young leaves of Arachis was established.The obtained DNA was detected by nucleic acid and protein analyzer, which results showed that A260/A280 ranged from 1.84 to 1.95, the extracted products had a bright band without tailing which could be observed through agarose gel electrophoresis. These results indicated that the genomic DNA being extracted by improved CTAB method had good integrality and high purity, and it could be used in each kinds of molecular markers research.
