摘要
目的观察锰对大鼠脑片神经细胞内质网应激信号分子的影响及其与细胞凋亡的关系。方法以胎大鼠脑片为模型,用400μLmol/L锰分别处理脑片6、12、18、24 h后,检测培养液中乳酸脱氢酶(LDH)的释放量,神经细胞凋亡率以及脑片中GRP78、CHOP及caspase 12的表达。结果随着锰处理时间的增加,脑片细胞损伤明显,LDH释放量,细胞凋亡率和GRP78、CHOP及caspase 12的表达均逐渐上升。结论锰可以时间依赖性地活化内质网应激信号分子,促使细胞凋亡。
Objective To study the effect of manganese (Mn) on the stress signaling moleculea of endoplasmic reticulum in the neural cells of rat brain slices and its relationship with apoptosis. Methods After the brain slices of the rats were respec- tively treated with 400/maol/L Mn for 6h, 12h, 18h and 24h, the lactate dehydrogenase (LDH) release in the culture medium, the neuronal apoptc^sis rate, and the expressions of GRP78, CHOP and caspase 12 in the brain slices were detected. Results With the increase of Mn treatment time, the cells in the brain slices were damaged significantly, and the release of LDH, the ap- optosis rate and expressions of GRP78, CHOP and easpase 12 were gradually increased. Conclusions Manganese can activate the stress signaling molecules of endoplasmic reticulum in the neural cells in a time - dependent manner and induces apoptosis.
