摘要
目的以乙醇和高脂膳食复合因素建立大鼠脂肪肝模型。方法将雌性Wistar大鼠随机分成阴性对照组、模型组,阴性对照组饲以普通饲料,自由饮水,每天以生理盐水灌胃;模型组饲以高脂饲料,饮用15%乙醇水,每天50%乙醇溶液灌胃;实验持续5周,测定大鼠血清中甘油三酯(TG)、总胆固醇(TC)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST),以及肝组织的TG、TC含量,并观察肝脏病理学变化。结果与阴性对照组比较,模型组大鼠肝指数有所升高,但差异无统计学意义(P>0.05);血清中TC明显升高[(2.93±0.62)mmol/L],与阴性对照组[(2.27±0.28)mmol/L]相比,差异有统计学意义(P<0.05);TG有所升高,但差异无统计学意义(P>0.05);肝组织中TG、TC明显升高(P<0.05);与阴性对照组比较,模型组大鼠血清中AST和ALT活力差异均无统计学意义(P>0.05);与阴性对照组比较,模型组大鼠的食物利用率明显减少(P<0.05);病理切片显示模型组肝脏发生明显的脂肪变性。结论乙醇加高脂膳食持续5周可以成功建立大鼠脂肪肝模型。
[ Objective] To establish a rat model of fatty liver based on alcohol and high fat diet. [ Methods] Female Wistar rats were randomly divided into the control group and the model group. Rats in the control group were fed by normal food and water, and given the normal saline via intragastric administration daily. Rats in the model group were feed by high fat food and 15% alcohol water, and given the 50% alcohol solution via intragastric administration daily. The experiment was lasting for 5 weeks. The levels of triglyceride (TG), total cholesterol (TC), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in serum, as well as TG and TC in hepatic tissue were measured. The pathological condition of liver was observed. [ Results] The index of liver in the model group was higher than that in the control group, but the difference was not significant ( P 〉 0.05 ). The serum TC level in the model group [ (2.93 ±0. 62 ) mmol/L] was higher than that in the control group [ [ 2.27 ± 0.28 ) mmol/L] significantly ( P 〈 0.05 ). The serum TG level increased, but the difference was not significant ( P 〉 0.05 ). The levels of TG and TC in hepatic tissue increased significantly (P 〈 0.05 ). There were no significant differences in serum AST and ALT activity between two groups ( P 〉 0.05 ). The food utilization rate in the model group was significantly lower than that in the control group ( P 〈 0.05 ). The pathological section showed that the obvious fatty degeneration was observed in hepatic tissue of rats in the model group. [ Conclusion] The fatty liver model of rat is successfully established by applying the alcohol and high fat diet for 5 weeks.
